The Study of PNIIIP in Sera of Normal Rats and Liver Cirrhosis Induced Rats




Abstract
The massive collagen deposition in portal triads and stroma means liver fibrosis and liver eirrhosis. We investigated the collagen content in liver tissue and circulating procollagen type III peptide (PNIIIP) in sera by quantitative measurement.
Experimental cirrhosis was induced in rat by using of bile duct ligation/scission operation(BDL/s). Serum PNIIIP values were measured with the ELISA. Hydroxyproline contents in the fibrotic livers was also examined by colorimetry. Serum
biochemical
tests were conducted to evaluate liver function.
Serum PNIIIP value increased and became highest at 4 wee4k of post-operation followed by maintaining the highest level in BDL/s model. The collagen content started to increase at 2 week and liver cirrhosis with bile duct hyperplasia at 4 week of
post-operation in BDL/s model(8.34¡¾11.4ng/ml, 53.62¡¾20.0ng/ml). Hydroxyproline content at 4 week of post-operation was 2~3 times higher than sham operated rats (84.9¡¾22.6§¶/g, 172.4¡¾66.8§¶/g). The significant correlation between PNIIIP in
sera
and
hydroxyproline in liver tissue 9r=0.878, p<0.005), but no correlation of clinical biochemistry in sera and of PNIIIP in sera or of hydroxyproline in liver tissue were observed. And the liver fibrosis(cirrhosis) showed at 3~4 week of
post-operation
by
light microscopy observation.
The results indicated that the detection of PNIIIP in sera and total collagen in liver tissue were reliable method in fibrotic or cirrhotic liver
